Pcr en tiempo real como herramienta de verificación de la presencia de salmonella spp. En un sistema haccp implementado en Cárnicos

Salmonella spp. en alimentos continua siendo un peligro microbiológico, que debe ser controlado en las plantas de producción de alimentos. Este estudio evaluó la presencia de Salmonella spp. en productos cárnicos de una planta con certificación HACCR usando PCR en tiempo real, como herramienta de verificación del sistema. Las muestras, se analizaron por microbiología tradicional y PCR TR. Los análisis microbiológicos, se realizaron siguiendo la metodología descrita por el INVIMA con identificación por API 20E. Se analizaron 67 muestras, 50 de producto terminado y 17 de materia prima. Con la metodología tradicional, se recuperaron 115 cepas sospechosas que fueron identificadas principalmente como Citrobacter spp., Hafnia aluei y Proleus spp. Se detecto presencia de Salmonella spp. en cuatro muestras de materia prima, en los correspondientes productos terminados no se encontró esta bacteria. PCR TR fue positivo para la presencia de Salmonella spp. en tres de las muestras mencionadas anteriormente. El crecimiento de microorganismos interferentes en los medios selectivos usados por el método tradicional, revela la baja selectividad de los mismos afectando la recuperación de Salmonella spp. y generar incertidumbre en los resultados. Finalmente, se puede concluir que la ausencia de Salmonella spp. en el producto terminado indica que el sistema HACCP ¡mplementado es efectivo y que PCR TR es una excelente alternativa como herramienta de verificación de los sistemas de calidad, ya que además de generar resultados rápidos tiene un coeficiente de correlación alto (0,86) con el método tradicional conocido como prueba Gold Estándar. In this study the presence of Salmonella spp. in meat producís from a plañí with a certified HACCP system was evaluated, using real time PCR as a verlfication tool. Samples were analyzed by classic microbiology technlques and real time PCR. For the traditional method the INVIMA procedure was followed and API 20E was used in the identification process. 67 samples were analyzed, 50 corresponding to final product and 17 to raw material. 115 suspicious strains were isolated by the traditional method, which were mainly identified as Citrobacter spp., Hafnia alvei and Proteus spp. The presence of Salmonella spp. in four samples of raw material was also detected by this method, no Salmonella spp were found in final product. Real time PCR was positive for the presence oí Salmonella spp. in three of the former samples. The growth of other microorganisms different from Salmonella spp. in the selective media, revealed low selectivity and low recovery of the microorganism. Finally it can be concluded that the absence of Salmonella spp. indicates that the implemented HACCP system is effective and that real time PCR is an excellent alternative as a verification tool for food safety systems. This because it generates fast results and also has a 0.86 correlation coefficient with the traditional method known as Gold Standard test.
