Estudio fitoquímico preliminar de la fracción polar de raíz de Zanthoxylum caribaeum (Rutaceae)

El material vegetal de Zanthoxylum caribaeum fue recolectado en el municipio de Apulo departamento Cundinamarca, una vez seco se sometió al proceso de extracción con etanol mediante maceración; y luego se realizó un fraccionamiento sólido-liquido por cromatografía líquida al vacío al extracto etanólico de la raíz. A partir de algunas de las fracciones obtenidas se realiza el presente estudio, las cuales fueron suministradas por el grupo de investigación en productos naturales Quipronab de la Universidad Nacional de Colombia. De esta manera, se realizó un estudio cromatográfico para separar y purificar algunos de los compuestos presentes en dichas fracciones, mediante cromatografía en capa fina (CCF), cromatografía en columna (CC) y cromatografía líquida al vacío (CLV). La identificación y elucidación estructural de los compuestos aislados se realizó por medio de reactivo de Dragendorff y técnicas espectroscópicas de RMN. En total se lograron identificar dos compuestos mayoritarios de las raíces de la especie Zanthoxylum caribaeum. Estos compuestos son de tipo alcaloidal, el cual uno de ellos se identificó como berberina y el otro fue identificado como el ácido 3-(9H- β-carbolin-1-il) propanoico, el cual mostró tener un efecto neuroprotector frente al neurotóxico paraquat en células SHSY5Y a concentraciones de 0.1 μg/mL. The plant material of Zanthoxylum caribaeum was collected in the municipality of Apulo, department of Cundinamarca, once dry it was subjected to the process of extraction with ethanol by maceration; and then a solid-liquid fractionation was carried out by vacuum liquid chromatography to the ethanolic extract of the root. The present study is carried out from some of the obtained fractions, which were supplied by the Quipronab natural products research group of the National University of Colombia. In this way, a chromatographic study was carried out to separate and purify some of the compounds present in said fractions, by thin layer chromatography (TLC), column chromatography (CC) and vacuum liquid chromatography (LCV). The identification and structural elucidation of the isolated compounds was carried out by means of the Dragendorff reagent and NMR spectroscopic techniques. In total, two major compounds from the roots of the Zanthoxylum caribaeum species were identified. These compounds are of the alkaloidal type, one of which was identified as berberine and the other was identified as 3- (9H- β-carbolin-1-yl) propanoic acid, which was shown to have a neuroprotective effect against the neurotoxic paraquat. in SHSY5Y cells at concentrations of 0.1 μg / mL.
